The Melba Highway, Castella is closed in both directions between Healesville-Kinglake Road and Old Toolangi Road following a serious collision.
Motorists should consider using the Maroondah Highway through Healesville, Narbethong, Buxton and Alexandra to Yarck, then the Goulburn Valley Highway back to Yea.
This is a significant detour which will add more than 100km to the journey, and extend travel times by up to two hours.
Heidelberg-Kinglake Road is a challenging road not suitable for all vehicles, but may provide a shorter detour around the closure through St Andrews and Kinglake.
Whittlesea-Kinglake Road provides a similar option.
The closure is under the control of Victoria Police.
The Melba Highway will reopen as soon as it is safe to do so.
